A stopped-flow method for the simultaneous determination of epinephrine and norepinephrine, two catecholamines with a wide spectrum of biological activity, is proposed. The method is based on the oxidation of these compounds with 1,10-phenanthroline-iron(III) complex, which is monitored by measuring the initial rate of change of the absorbance of the ferroin formed at 510 nm. The difference in kinetic behaviour between the two species was exploited by applying a modified version of the proportional-equation method for the resolution of epinephrine-norepinephrine mixtures at the micrograms ml-1 level over the ratio range 1:10-10:1, with an error of less than 5% and an average precision of about 2.5%. The method was successfully applied to the determination of the two catecholamines in pharmaceuticals.

UI MeSH Term Description Entries
D007700 Kinetics The rate dynamics in chemical or physical systems.
D009638 Norepinephrine Precursor of epinephrine that is secreted by the ADRENAL MEDULLA and is a widespread central and autonomic neurotransmitter. Norepinephrine is the principal transmitter of most postganglionic sympathetic fibers, and of the diffuse projection system in the brain that arises from the LOCUS CERULEUS. It is also found in plants and is used pharmacologically as a sympathomimetic. D004837 Epinephrine The active sympathomimetic hormone from the ADRENAL MEDULLA. It stimulates both the alpha- and beta- adrenergic systems, causes systemic VASOCONSTRICTION and gastrointestinal relaxation, stimulates the HEART, and dilates BRONCHI and cerebral vessels. It is used in ASTHMA and CARDIAC FAILURE and to delay absorption of local ANESTHETICS. D013053 Spectrophotometry The art or process of comparing photometrically the relative intensities of the light in different parts of the spectrum.
