In in vitro experiments, amiridine in concentration of 100, 200, and 300 microM restored disturbed structure of microtubules assembled from tubulin and microtubule-associated proteins isolated from the brain of patients with Alzheimer disease. Tacrine in a concentration of 100, 250, and 500 microM inhibited phosphorylation of tubulin and microtubule-associated proteins isolated from the brain of patients with Alzheimer disease, while memantine and amiridine in the specified concentrations had no effect on phosphorylation.

UI MeSH Term Description Entries
D008559 Memantine AMANTADINE derivative that has some dopaminergic effects. It has been proposed as an antiparkinson agent. 1,3-Dimethyl-5-aminoadamantane,1-Amino-3,5-dimethyladamantane,Axura,D-145,D145,Ebixa,Memantin,Memantine Hydrochloride,Namenda,D 145
D008869 Microtubule-Associated Proteins High molecular weight proteins found in the MICROTUBULES of the cytoskeletal system. Under certain conditions they are required for TUBULIN assembly into the microtubules and stabilize the assembled microtubules. D008870 Microtubules Slender, cylindrical filaments found in the cytoskeleton of plant and animal cells. They are composed of the protein TUBULIN and are influenced by TUBULIN MODULATORS. Microtubule
D010766 Phosphorylation The introduction of a phosphoryl group into a compound through the formation of an ester bond between the compound and a phosphorus moiety. Phosphorylations
D001921 Brain The part of CENTRAL NERVOUS SYSTEM that is contained within the skull (CRANIUM). Arising from the NEURAL TUBE, the embryonic brain is comprised of three major parts including PROSENCEPHALON (the forebrain); MESENCEPHALON (the midbrain); and RHOMBENCEPHALON (the hindbrain). The developed brain consists of CEREBRUM; CEREBELLUM; and other structures in the BRAIN STEM. Encephalon

D000544 Alzheimer Disease A degenerative disease of the BRAIN characterized by the insidious onset of DEMENTIA. Impairment of MEMORY, judgment, attention span, and problem solving skills are followed by severe APRAXIAS and a global loss of cognitive abilities. The condition primarily occurs after age 60, and is marked pathologically by severe cortical atrophy and the triad of SENILE PLAQUES; NEUROFIBRILLARY TANGLES; and NEUROPIL THREADS. D000634 Aminoquinolines Quinolines substituted in any position by one or more amino groups.
D013619 Tacrine A cholinesterase inhibitor that crosses the blood-brain barrier. Tacrine has been used to counter the effects of muscle relaxants, as a respiratory stimulant, and in the treatment of Alzheimer's disease and other central nervous system disorders. D014229 Triazolam A short-acting benzodiazepine used in the treatment of insomnia. Some countries temporarily withdrew triazolam from the market because of concerns about adverse reactions, mostly psychological, associated with higher dose ranges. Its use at lower doses with appropriate care and labeling has been reaffirmed by the FDA and most other countries.
