Intense recent effort towards the development of new drug candidates in the macrolide arena has resulted in the emergence of a number of novel chemical entities; two of these, ABT-773 and HMR-3647, are currently at the advanced clinical trial stage. This review covers the various approaches described in the literature specifically for the synthesis of the ketolide class of compounds.
