By far the most convenient and simple assays for screening monoclonal antibody-producing hybridomas are those utilizing solid-phase binding assays, where radiolabelled or enzyme linked antispecies antibodies are used to detect binding of the hybridoma supernatants to insolubilized antigen. Such procedures require an adequate supply of antigen, preferably purified, which can be bound to a solid support, usually through noncovalent binding to PVC microtiter plates or nitrocellulose sheets.
