BACKGROUND Immunolocalization of cellular antigens typically requires fixation and permeabilization of cells, prior to incubation with antibodies. RESULTS Assessing a test protein abundantly present at the cell surface of Dictyostelium cells, we show that in fixed cells, permeabilization extracts almost completely this cell surface antigen. The extent of this artifact is variable depending on the procedure used for labeling and permeabilization, as well as on the antigen considered. CONCLUSIONS An optimized protocol for labeling both surface and intracellular antigens without significant loss of labeling is proposed.

UI MeSH Term Description Entries
D004023 Dictyostelium A genus of protozoa, formerly also considered a fungus. Its natural habitat is decaying forest leaves, where it feeds on bacteria. D. discoideum is the best-known species and is widely used in biomedical research. Dictyostelium discoideum,Dictyostelium discoideums,Dictyosteliums, discoideum, Dictyostelium
D005455 Fluorescent Antibody Technique Test for tissue antigen using either a direct method, by conjugation of antibody with fluorescent dye (FLUORESCENT ANTIBODY TECHNIQUE, DIRECT) or an indirect method, by formation of antigen-antibody complex which is then labeled with fluorescein-conjugated anti-immunoglobulin antibody (FLUORESCENT ANTIBODY TECHNIQUE, INDIRECT). The tissue is then examined by fluorescence microscopy.
