The use of achiral/chiral coupled-column HPLC in the direct analysis of the enantiomers of chiral drugs in biological samples is reviewed. The method has been applied to a study of the pharmacokinetics of (+)- and (-)-terbutaline, the resolution of the stereoisomers of leucovorin and the determination of warfarin enantiomers in serum.
