Hypersensitivity reactions to etoposide. A case report and review of the literature. 1988

A case of immediate hypersensitivity to the chemotherapeutic agent etoposide is reported. The clinical details of the case and features of previously reported cases are outlined. An attempt to re-treat the patient with etoposide was unsuccessful despite vigorous premedication, in contrast to previous reports. The mechanism of hypersensitivity to this agent is still to be elucidated.

D005047 Etoposide A semisynthetic derivative of PODOPHYLLOTOXIN that exhibits antitumor activity. Etoposide inhibits DNA synthesis by forming a complex with topoisomerase II and DNA. This complex induces breaks in double stranded DNA and prevents repair by topoisomerase II binding. Accumulated breaks in DNA prevent entry into the mitotic phase of cell division, and lead to cell death. Etoposide acts primarily in the G2 and S phases of the cell cycle.
