Quantifying the titer of African swine fever virus is critical for disease control, viral infection studies, and disinfectant efficacy tests. Techniques such as real-time PCR and virus isolation provide an understanding as to whether viral genome is present or gives a qualitative assessment of live viral presence in a sample respectively, but neither provide a quantitative measurement of live virus. Here we describe a plaque assay for the titration of a Vero-adapted African swine fever virus strain (BA71V) and describe how to apply this method to determine disinfectant efficacy.

UI MeSH Term Description Entries
D004202 Disinfectants Substances used on inanimate objects that destroy harmful microorganisms or inhibit their activity. Disinfectants are classed as complete, destroying SPORES as well as vegetative forms of microorganisms, or incomplete, destroying only vegetative forms of the organisms. They are distinguished from ANTISEPTICS, which are local anti-infective agents used on humans and other animals. (From Hawley's Condensed Chemical Dictionary, 11th ed) Biocide,Disinfectant,Biocides
D004267 DNA Viruses Viruses whose nucleic acid is DNA. DNA Virus,Virus, DNA,Viruses, DNA
D000357 African Swine Fever A sometimes fatal ASFIVIRUS infection of pigs, characterized by fever, cough, diarrhea, hemorrhagic lymph nodes, and edema of the gallbladder. It is transmitted between domestic swine by direct contact, ingestion of infected meat, or fomites, or mechanically by biting flies or soft ticks (genus Ornithodoros). D000358 African Swine Fever Virus The lone species of the genus Asfivirus. It infects domestic and wild pigs, warthogs, and bushpigs. Disease is endemic in domestic swine in many African countries and Sardinia. Soft ticks of the genus Ornithodoros are also infected and act as vectors. D000818 Animals Unicellular or multicellular, heterotrophic organisms, that have sensation and the power of voluntary movement. Under the older five kingdom paradigm, Animalia was one of the kingdoms. Under the modern three domain model, Animalia represents one of the many groups in the domain EUKARYOTA. D013552 Swine Any of various animals that constitute the family Suidae and comprise stout-bodied, short-legged omnivorous mammals with thick skin, usually covered with coarse bristles, a rather long mobile snout, and small tail. Included are the genera Babyrousa, Phacochoerus (wart hogs), and Sus, the latter containing the domestic pig (see SUS SCROFA).
