Preparation of ultrathin frozen sections in electron microscopy: specimen supported by polyethylene glycol. 1977

M Novák, and A Jelínková, and R Benda

A method was worked out for supporting biological specimens with polyethylene glycol (PEG) for the preparation of ultrathin frozen sections. This method was used in the preparation of ultra-thin cryosections of various tissues and of human diploid cell suspensions infected with vaccina and Herpes simplex viruses. In such preparations it was possible to study stages of virus particle formation under the electron microscope.

UI MeSH Term Description Entries
D007668 Kidney Body organ that filters blood for the secretion of URINE and that regulates ion concentrations. Kidneys
D008854 Microscopy, Electron Microscopy using an electron beam, instead of light, to visualize the sample, thereby allowing much greater magnification. The interactions of ELECTRONS with specimens are used to provide information about the fine structure of that specimen. In TRANSMISSION ELECTRON MICROSCOPY the reactions of the electrons that are transmitted through the specimen are imaged. In SCANNING ELECTRON MICROSCOPY an electron beam falls at a non-normal angle on the specimen and the image is derived from the reactions occurring above the plane of the specimen. Electron Microscopy
D008867 Microtomy The technique of using a microtome to cut thin or ultrathin sections of tissues embedded in a supporting substance. The microtome is an instrument that hold a steel, glass or diamond knife in clamps at an angle to the blocks of prepared tissues, which it cuts in sections of equal thickness. D011092 Polyethylene Glycols Polymers of ETHYLENE OXIDE and water, and their ethers. They vary in consistency from liquid to solid depending on the molecular weight indicated by a number following the name. They are used as SURFACTANTS, dispersing agents, solvents, ointment and suppository bases, vehicles, and tablet excipients. Some specific groups are NONOXYNOLS, OCTOXYNOLS, and POLOXAMERS. D006564 Herpesviridae A family of enveloped, linear, double-stranded DNA viruses infecting a wide variety of animals.
