Complement fixation antibodies to glycosphingolipids in sera of rare blood group p and Pk phenotypes. 1978

M Kato, and S Kubo, and M Naiki

Blood group P antigen and Pk antigen were recently identified as glycosphingolipid globoside, GalNAc(beta, 1 leads to 3)Gal(alpha, 1 leads to 4)Gas(beta, 1 leads to 4)Glc(beta, 1 leads to 1)ceramide, and trihexosylceramide, Gal(alpha, 1 leads to 4)Gal(beta, 1 leads to 4)Gld(beta, 1 leads to 1)ceramide, respectively (Naiki & Marcus, 1974). The presence of antibodies to both antigens in the sera from rare p donors and antibodies to P antigen in the serum from a rare Pk donor were first demonstrated by the complement fixation test with the optimal antigen preparations containing each purified glycoplipid, lecithin and cholesterol. In the serum from a p donor, most of antibody population of anti-P was IgM and was cross-reacted to Forssman glycolipid, GalNAc(alpha, 1 leads to 3)GalNAc(beta, 1 leads to 3)Gal(alpha, 1 leads to 4)Gal(beta, 1 leads to 4)Glc(beta 1 leads to 1) ceramide, but the rest of the anti-P antibodies and most of anti-Pk antibodies were IgG and had marked specificities for globoside and trihexosylceramide, respectively.

UI MeSH Term Description Entries
D007074 Immunoglobulin G The major immunoglobulin isotype class in normal human serum. There are several isotype subclasses of IgG, for example, IgG1, IgG2A, and IgG2B. D007075 Immunoglobulin M A class of immunoglobulin bearing mu chains (IMMUNOGLOBULIN MU-CHAINS). IgM can fix COMPLEMENT. The name comes from its high molecular weight and originally was called a macroglobulin. Gamma Globulin, 19S,IgM,IgM Antibody,IgM1,IgM2,19S Gamma Globulin,Antibody, IgM
D010127 P Blood-Group System A blood group related to the ABO, Lewis and I systems. At least five different erythrocyte antigens are possible, some very rare, others almost universal. Multiple alleles are involved in this blood group. Blood-Group System, P,Blood-Group Systems, P,P Blood Group System,P Blood-Group Systems,System, P Blood-Group,Systems, P Blood-Group
D010641 Phenotype The outward appearance of the individual. It is the product of interactions between genes, and between the GENOTYPE and the environment. Phenotypes
D001789 Blood Group Antigens Sets of cell surface antigens located on BLOOD CELLS. They are usually membrane GLYCOPROTEINS or GLYCOLIPIDS that are antigenically distinguished by their carbohydrate moieties. D003168 Complement Fixation Tests Serologic tests based on inactivation of complement by the antigen-antibody complex (stage 1). Binding of free complement can be visualized by addition of a second antigen-antibody system such as red cells and appropriate red cell antibody (hemolysin) requiring complement for its completion (stage 2). Failure of the red cells to lyse indicates that a specific antigen-antibody reaction has taken place in stage 1. If red cells lyse, free complement is present indicating no antigen-antibody reaction occurred in stage 1. D006028 Glycosphingolipids Lipids containing at least one monosaccharide residue and either a sphingoid or a ceramide (CERAMIDES). They are subdivided into NEUTRAL GLYCOSPHINGOLIPIDS comprising monoglycosyl- and oligoglycosylsphingoids and monoglycosyl- and oligoglycosylceramides; and ACIDIC GLYCOSPHINGOLIPIDS which comprises sialosylglycosylsphingolipids (GANGLIOSIDES); SULFOGLYCOSPHINGOLIPIDS (formerly known as sulfatides), glycuronoglycosphingolipids, and phospho- and phosphonoglycosphingolipids. D006801 Humans Members of the species Homo sapiens. Homo sapiens,Man (Taxonomy),Human,Man, Modern,Modern Man
D000906 Antibodies Immunoglobulin molecules having a specific amino acid sequence by virtue of which they interact only with the ANTIGEN (or a very similar shape) that induced their synthesis in cells of the lymphoid series (especially PLASMA CELLS).
D000918 Antibody Specificity The property of antibodies which enables them to react with some ANTIGENIC DETERMINANTS and not with others. Specificity is dependent on chemical composition, physical forces, and molecular structure at the binding site. Antibody Specificities,Specificities, Antibody,Specificity, Antibody
