[Radioligand assay: method and application. II. Effect of glucagon coupling on the production of glucagon antisera with suitable binding parameters for radioimmunoassay]. 1980

Glucagon antibodies were produced in rabbits using three immunogenic glucagon conjugates. Glucagon was coupled to bovine serum albumin by difluorodinitrobenzene (DFDNB), carbodiimide (ECDI) or glutardialdehyde. Rabbits immunized with glucagon conjugated to albumin using DFDNB produced sensitive antisera for radioimmunoassay quite specific for pancreatic glucagon. The affinity constant of the best antiserum was approximately 10(11) l/mol. Antisera raised against the two other glucagon-conjugates were significantly less affine. All of these antisera showed inverse binding curves of 126J-glucagon caused by positive cooperatively in dependence upon the antigen/antibody ratio. The race of rabbits used for immunization was without influence on the immune response.

UI MeSH Term Description Entries
D007106 Immune Sera Serum that contains antibodies. It is obtained from an animal that has been immunized either by ANTIGEN injection or infection with microorganisms containing the antigen. Antisera,Immune Serums,Sera, Immune,Serums, Immune
D011485 Protein Binding The process in which substances, either endogenous or exogenous, bind to proteins, peptides, enzymes, protein precursors, or allied compounds. Specific protein-binding measures are often used as assays in diagnostic assessments. Plasma Protein Binding Capacity,Binding, Protein
D011817 Rabbits A burrowing plant-eating mammal with hind limbs that are longer than its fore limbs. It belongs to the family Leporidae of the order Lagomorpha, and in contrast to hares, possesses 22 instead of 24 pairs of chromosomes. D011863 Radioimmunoassay Classic quantitative assay for detection of antigen-antibody reactions using a radioactively labeled substance (radioligand) either directly or indirectly to measure the binding of the unlabeled substance to a specific antibody or other receptor system. Non-immunogenic substances (e.g., haptens) can be measured if coupled to larger carrier proteins (e.g., bovine gamma-globulin or human serum albumin) capable of inducing antibody formation. Radioimmunoassays
D002234 Carbodiimides Compounds with the general formula RN Carbodiimide
D004139 Dinitrofluorobenzene Irritants and reagents for labeling terminal amino acid groups. DNFB,Fluorodinitrobenzene,1-Fluoro-2,4-dinitrobenzene,1 Fluoro 2,4 dinitrobenzene
D005934 Glucagon A 29-amino acid pancreatic peptide derived from proglucagon which is also the precursor of intestinal GLUCAGON-LIKE PEPTIDES. Glucagon is secreted by PANCREATIC ALPHA CELLS and plays an important role in regulation of BLOOD GLUCOSE concentration, ketone metabolism, and several other biochemical and physiological processes. D000818 Animals Unicellular or multicellular, heterotrophic organisms, that have sensation and the power of voluntary movement. Under the older five kingdom paradigm, Animalia was one of the kingdoms. Under the modern three domain model, Animalia represents one of the many groups in the domain EUKARYOTA.
