Basic proline-rich proteins from human parotid saliva: complete covalent structure of protein IB-9 and partial structure of protein IB-6, members of a polymorphic pair. 1982

D Kauffman, and R Wong, and A Bennick, and P Keller

The complete amino acid sequence of a basic proline-rich protein, IB-9, from human parotid saliva was determined by automated Edman degradation of peptides obtained by enzymatic cleavage of the intact protein with clostripain. The protein was digested with papain and elastase to obtain overlapping peptides. Automated Edman degradation of the intact protein was also performed. The protein consists of 61 amino acids, of which 26 are proline residues. The partial sequence of another human parotid basic proline-rich protein, IB-6, was also obtained. With one exception the first 54 residues of the two proteins are identical. An exceptional degree of internal reiteration occurs in both molecules, including several repeated sequences of 12-14 amino acids. The proteins show a high degree of homology with the C-terminal portion of the salivary acidic proline-rich protein C.

UI MeSH Term Description Entries
D010196 Pancreatic Elastase A protease of broad specificity, obtained from dried pancreas. Molecular weight is approximately 25,000. The enzyme breaks down elastin, the specific protein of elastic fibers, and digests other proteins such as fibrin, hemoglobin, and albumin. EC 3.4.21.36. D010206 Papain A proteolytic enzyme obtained from Carica papaya. It is also the name used for a purified mixture of papain and CHYMOPAPAIN that is used as a topical enzymatic debriding agent. EC 3.4.22.2. Tromasin
D010306 Parotid Gland The largest of the three pairs of SALIVARY GLANDS. They lie on the sides of the FACE immediately below and in front of the EAR. Gland, Parotid,Glands, Parotid,Parotid Glands
D010455 Peptides Members of the class of compounds composed of AMINO ACIDS joined together by peptide bonds between adjacent amino acids into linear, branched or cyclical structures. OLIGOPEPTIDES are composed of approximately 2-12 amino acids. Polypeptides are composed of approximately 13 or more amino acids. PROTEINS are considered to be larger versions of peptides that can form into complex structures such as ENZYMES and RECEPTORS. Peptide,Polypeptide,Polypeptides
D011392 Proline A non-essential amino acid that is synthesized from GLUTAMIC ACID. It is an essential component of COLLAGEN and is important for proper functioning of joints and tendons. L-Proline,L Proline
D000595 Amino Acid Sequence The order of amino acids as they occur in a polypeptide chain. This is referred to as the primary structure of proteins. It is of fundamental importance in determining PROTEIN CONFORMATION. D012463 Saliva The clear, viscous fluid secreted by the SALIVARY GLANDS and mucous glands of the mouth. It contains MUCINS, water, organic salts, and ptylin. Salivas
D012471 Salivary Proteins and Peptides Proteins and peptides found in SALIVA and the SALIVARY GLANDS. Some salivary proteins such as ALPHA-AMYLASES are enzymes, but their composition varies in different individuals. D055232 Proline-Rich Protein Domains Protein domains that are enriched in PROLINE. The cyclical nature of proline causes the peptide bonds it forms to have a limited degree of conformational mobility. Therefore the presence of multiple prolines in close proximity to each other can convey a distinct conformational arrangement to a peptide chain. D055273 Salivary Proline-Rich Proteins A family of proline-rich proteins that constitute the majority of the protein component of SALIVA. Salivary proline-rich proteins occur as acidic, basic and glycosylated basic proteins. They perform a variety of functions such as adhering to the acquired ENAMEL PELLICLE, acting as lubricants and precipitating TANNINS.
