Isolation of the newly synthesized DNA of HeLa cells containing beta-2'-deoxy-6-thioguanylate on an organomercurial agarose column. 1980

S Yoshida, and T Morita, and M Saneyoshi

It has been shown that beta-2'-deoxy-6-thioguanosine is incorporated into mammalian DNA via its triphosphate form. The newly synthesized DNA of HeLa cells containing thioguanosine, adsorbed specifically on an organomercurial agarose column and was eluted with 2-mercaptoethanol. By recycling this affinity chromatography, the newly synthesized DNA strands were separated successfully from non-adsorbable parental strands. This system may provide a new tool for the study of DNA replication.

UI MeSH Term Description Entries
D009941 Organomercury Compounds Organic compounds which contain mercury as an integral part of the molecule. Compounds, Organomercury
D002846 Chromatography, Affinity A chromatographic technique that utilizes the ability of biological molecules, often ANTIBODIES, to bind to certain ligands specifically and reversibly. It is used in protein biochemistry. (McGraw-Hill Dictionary of Scientific and Technical Terms, 4th ed) Chromatography, Bioaffinity,Immunochromatography,Affinity Chromatography,Bioaffinity Chromatography
D003848 Deoxyguanine Nucleotides Guanine nucleotides which contain deoxyribose as the sugar moiety. Deoxyguanosine Phosphates,Nucleotides, Deoxyguanine,Phosphates, Deoxyguanosine
D004247 DNA A deoxyribonucleotide polymer that is the primary genetic material of all cells. Eukaryotic and prokaryotic organisms normally contain DNA in a double-stranded state, yet several important biological processes transiently involve single-stranded regions. DNA, which consists of a polysugar-phosphate backbone possessing projections of purines (adenine and guanine) and pyrimidines (thymine and cytosine), forms a double helix that is held together by hydrogen bonds between these purines and pyrimidines (adenine to thymine and guanine to cytosine). DNA, Double-Stranded,Deoxyribonucleic Acid,ds-DNA,DNA, Double Stranded,Double-Stranded DNA,ds DNA
D006367 HeLa Cells The first continuously cultured human malignant CELL LINE, derived from the cervical carcinoma of Henrietta Lacks. These cells are used for, among other things, VIRUS CULTIVATION and PRECLINICAL DRUG EVALUATION assays. Cell, HeLa,Cells, HeLa,HeLa Cell
D006801 Humans Members of the species Homo sapiens. Homo sapiens,Man (Taxonomy),Human,Man, Modern,Modern Man
D013010 Sonication The application of high intensity ultrasound to liquids. Sonications
D013873 Thionucleotides Nucleotides in which the base moiety is substituted with one or more sulfur atoms.
