Effects of short and long term administrations of an oral contraceptive preparation on the pituitary responses to the synthetic LH-RH were compared in the present experiment. Five normal cycling women were used as controls in the late proliferative phase. Five healty volunteers taking an oral contraceptive agent (1.0 mg norethindrone with 0.05 mg mestranol) for less than 10 cycles were served as short term group and other 4 women taking the same preparation for more than 40 cycles were studied as long term group. Volunteers in each group were kept fasting overnight and 5 ml of blood was drawn at 9 to 10 am on the day of experiment. Ten minutes after drawing blood, 200 mug synthetic LH-RH was injected subcutaneously in the late proliferative phase in the normal cycling women. According to the same procedure, women taking pills were administered LH-RH on the 7th to 12th day after the beginning of taking the first tablet in each cyle. Three ml of blood was taken at 15, 30, 60, 120 minutes and 24 hours, and serum LH and Fsh were determined by the double antibody radioimmunoassay. The 2nd IRP-HMG was used as the standard materials and expressed as mIU/ml of serum. Mean baseline serum LH and FSH concentrations were not suppressed in a short term administration of norethindrone-mestranol combination. Fifteen minutes after the subcutaneous injection of LH-RH, mean LH level was significantly elevated, and thereafter the level was not significantly changed as compared with that seen in the late proliferative phase of the cycle. Concerning the FSH response to LH-RH, a short term administration did not induce a significant rise. ...

UI MeSH Term Description Entries
D007986 Luteinizing Hormone A major gonadotropin secreted by the adenohypophysis (PITUITARY GLAND, ANTERIOR). Luteinizing hormone regulates steroid production by the interstitial cells of the TESTIS and the OVARY. The preovulatory LUTEINIZING HORMONE surge in females induces OVULATION, and subsequent LUTEINIZATION of the follicle. LUTEINIZING HORMONE consists of two noncovalently linked subunits, alpha and beta. Within a species, the alpha subunit is common in the three pituitary glycoprotein hormones (TSH, LH and FSH), but the beta subunit is unique and confers its biological specificity. D007987 Gonadotropin-Releasing Hormone A decapeptide that stimulates the synthesis and secretion of both pituitary gonadotropins, LUTEINIZING HORMONE and FOLLICLE STIMULATING HORMONE. GnRH is produced by neurons in the septum PREOPTIC AREA of the HYPOTHALAMUS and released into the pituitary portal blood, leading to stimulation of GONADOTROPHS in the ANTERIOR PITUITARY GLAND. D008656 Mestranol The 3-methyl ether of ETHINYL ESTRADIOL. It must be demethylated to be biologically active. It is used as the estrogen component of many combination ORAL CONTRACEPTIVES. 19-Norpregna-1,3,5(10)-trien-20-yn-17-ol, 3-methoxy-, (17alpha)-,Ethinyl Estradiol 3-Methyl Ether,Ethinyl Estradiol 3 Methyl Ether
D009640 Norethindrone A synthetic progestational hormone with actions similar to those of PROGESTERONE but functioning as a more potent inhibitor of ovulation. It has weak estrogenic and androgenic properties. The hormone has been used in treating amenorrhea, functional uterine bleeding, endometriosis, and for CONTRACEPTION. 19-Norpregn-4-en-20-yn-3-one, 17-hydroxy-, (17alpha)-,Ethinylnortestosterone,Norethisterone,Norpregneninolone,Conceplan,Micronor,Monogest,Nor-QD,Norcolut,Norcolute,Norethindrone, (1 beta)-Isomer,Norlutin,Nor QD,NorQD
D010902 Pituitary Gland A small, unpaired gland situated in the SELLA TURCICA. It is connected to the HYPOTHALAMUS by a short stalk which is called the INFUNDIBULUM. D003280 Contraceptives, Oral, Synthetic Oral contraceptives which owe their effectiveness to synthetic preparations.
D005640 Follicle Stimulating Hormone A major gonadotropin secreted by the adenohypophysis (PITUITARY GLAND, ANTERIOR). Follicle-stimulating hormone stimulates GAMETOGENESIS and the supporting cells such as the ovarian GRANULOSA CELLS, the testicular SERTOLI CELLS, and LEYDIG CELLS. FSH consists of two noncovalently linked subunits, alpha and beta. Within a species, the alpha subunit is common in the three pituitary glycoprotein hormones (TSH, LH, and FSH), but the beta subunit is unique and confers its biological specificity.
