Increased phenytoin metabolism in rabbits after phenytoin pretreatment. 1976

The serum half-life of phenytoin in rabbits after a single intravenous dose was significantly shortened by pretreating the rabbits with daily oral doses of phenytoin. This suggests that in vivo phenytoin induces its own metabolism.

The mechanism of therapeutic action is not clear, although several cellular actions have been described including effects on ion channels, active transport, and general membrane stabilization.
