Assay of human rotavirus antibody by immune adherence hemagglutination with a cultivable human rotavirus as antigen. 1982

A cultivable human rotavirus, Wa, was propagated in MA-104 cells and used as an antigen in immune adherence hemagglutination (IAHA) and complement fixation (CF) tests. IAHA antibody titers of sera from normal humans were found to be eightfold higher than CF antibody titers. The IAHA antibody determination with the Wa antigen is considered to be especially useful for the seroepidemiological studies of human rotavirus infections.

Binding of free complement can be visualized by addition of a second antigen-antibody system such as red cells and appropriate red cell antibody (hemolysin) requiring complement for its completion (stage 2). Failure of the red cells to lyse indicates that a specific antigen-antibody reaction has taken place in stage 1. If red cells lyse, free complement is present indicating no antigen-antibody reaction occurred in stage 1.
